Reconstitution of Lyophilized Research Peptides

Lyophilized (freeze-dried) peptides arrive as a stable powder or cake. Correct reconstitution is essential to preserve the compound and to keep your experimental concentrations accurate. The following describes general laboratory technique for research handling only.

1. Equilibrate Before Opening

Allow the sealed vial to reach room temperature before breaking the seal. Opening a cold vial exposes the hygroscopic powder to condensation, which can introduce water and degrade sensitive sequences.

2. Select an Appropriate Solvent

Most peptides reconstitute in sterile water or bacteriostatic water for research applications. Highly hydrophobic sequences may require a small volume of a co-solvent (such as dilute acetic acid) before dilution. Always consult the solubility notes on the product page.

3. Add Solvent Slowly

Direct the solvent stream against the glass wall of the vial rather than onto the powder directly. Let the peptide dissolve passively. Avoid vigorous shaking, which can shear peptide bonds and cause foaming; gentle swirling is sufficient.

4. Calculate Concentration

Divide the net peptide mass by the total solvent volume to determine your working concentration. Account for net peptide content when high accuracy is required for quantitative work.

5. Aliquot and Store

Once reconstituted, divide the solution into single-use aliquots to avoid repeated freeze-thaw cycles, which are a leading cause of peptide degradation. Label each aliquot with the lot number and date.
